Tumorgenicity of different CTC morphometric subgroups was tested in vivo using mammary fat pad (MFP) implantation. All animal experiments were carried out in accordance with approved protocols from the Institutional Animal Care and Use Committee of USC. GFP-LUC-labeled BRx68 CTCs (BRx68_luc) were FACS sorted into three morphometric subgroups as described above. Tumors were established by inoculation of 200 sorted cells in 100 μL of Matrigel containing PBS into the 4th MFP of 6–8-week-old female NOD/SCID-Gamma (NSG) mice supplemented with subcutaneous slow-release estrogen pills. Tumor growth was monitored every 2 weeks by in vivo Bioluminescence imaging using IVIS Lumina III (PerkinElmer, Waltham, MA, USA) following intraperitoneal injection of 100 μL of d-luciferin substrate (Syd Labs, Hopkinton, MA, USA).

Cf2Th-CD4+ CCR5+ cells were seeded in 96-well luminometer-compatible plates at a density of 2 × 104 cells per well and infected the next day. For neutralization assays, virus preparations were incubated with the indicated concentration of the MAbs for 1 h at 37°C. In all neutralization assays, sample input was adjusted so that infectivity of any two variants did not vary by a factor of more than 2-fold. Samples were then added to Cf2Th-CD4+ CCR5+ cells and incubated for 3 days to allow infection. To measure infection, the medium was removed, and the cells were lysed with 35 μL passive lysis buffer (Promega) and subjected to three freeze-thaw cycles. To measure luciferase activity, 100 μL of luciferin buffer (15 mM MgSO4, 15 mM KPO4 [pH 7.6], 1 mM ATP, and 1 mM dithiothreitol) and 50 μL of 1 mM d-luciferin potassium salt (Syd Labs, MA) were added to each sample. Luminescence was recorded using a Synergy H1 microplate reader (BioTek Instruments).
