“Effector CD8+ T cells are important mediators of adaptive immunity, and receptor-ligand interactions that regulate their survival may have therapeutic potential. Here, we identified a subset of effector CD8+ T cells that expressed the inhibitory Fc receptor FcγRIIB following activation and multiple rounds of division. CD8+ T cell-intrinsic genetic deletion of Fcgr2b increased CD8+ effector T cell accumulation, resulting in accelerated graft rejection and decreased tumor volume in mouse models. IgG antibody was not required for FcγRIIB-mediated control of CD8+ T cell immunity, and instead, the immunosuppressive cytokine Fgl2 was a functional ligand for FcγRIIB on CD8+ T cells, in that Fgl2 induced caspase 3/7-mediated apoptosis in Fcgr2b+ but not Fcgr2b−/− CD8+ T cells. Moreover, Increased expression of FcγRIIB correlated with freedom from rejection following withdrawal from immunosuppression in a clinical trial of kidney transplant recipients. Together, these findings demonstrate a cell-intrinsic coinhibitory function of FcγRIIB in regulating CD8+ T cell immunity.”
2. Accelerated clearance and degradation of cell free HIV by neutralizing antibodies occurs via FcγRIIb on liver sinusoidal endothelial cells by endocytosis

“Neutralizing antibodies suppress HIV infection by accelerating viral clearance from blood circulation, in addition to neutralization. The elimination mechanism is largely unknown. We determined that human liver sinusoidal endothelial cells (LSEC) express FcγRIIb as the lone Fc gamma receptor, and, using humanized FcγRIIb mouse, we found that antibody-opsonized HIV pseudoviruses (Ab-HIV) were cleared considerably faster from circulation than HIV by LSEC FcγRIIb. Compared to humanized FcγRIIb expressing mice, HIV clearance was significantly slower in FcγRIIb KO mice. Interestingly, pentamix of neutralizing antibodies cleared HIV faster compared to polyclonal HIVIG, though the HIV antibody-antigen ratio was higher in immune complexes made of HIVIG and HIV than pentamix and HIV. The effector mechanism of LSEC FcγRIIb was identified to be endocytosis. Once endocytosed, both Ab-HIV and HIV localized to lysosomes. This suggests that clearance of HIV, endocytosis and lysosomal trafficking within LSEC occur sequentially, and that the clearance rate may influence downstream events. Most importantly, we have identified LSEC FcγRIIb-mediated endocytosis to be the Fc effector mechanism to eliminate cell free HIV by antibodies, which could inform development of HIV vaccine and antibody therapy.”
3. Plasmacytoid Dendritic Cells Take Up Opsonized Antigen Leading to CD4+ and CD8+ T Cell Activation In Vivo

“Plasmacytoid dendritic cells (pDC) are the body’s main source of IFN-α, but, unlike classical myeloid DC (myDC), they lack phagocytic activity and are generally perceived as playing only a minor role in Ag processing and presentation. We show that murine pDC, as well as myDC, express Fcγ receptors (CD16/CD32) and can use these receptors to acquire Ag from immune complexes (IC), resulting in the induction of robust Ag-specific CD4+ and CD8+ T cell responses. IC-loaded pDC stimulate CD4 + T cells to proliferate and secrete a mixture of IL-4 and IFN-γ, and they induce CD8+ T cells to secrete IL-10 as well as IFN-γ. In contrast, IC-loaded myDC induce both CD4+ and CD8+ T cells to secrete mainly IFN-γ. These results indicate that pDC can shape an immune response by acquiring and processing opsonized Ag, leading to a predominantly Th2 response.”
