Purified B5-D3 and control proteins were serially diluted and incubated with 50 PFU of SARS-CoV-2 (Wuhan-Hu-1, Delta, Omicron BA.5, BQ.1.22, and XBB.1.5) for 1 hr at room temperature, followed by addition to Vero E6 cells seeded in 6-well plates. After incubation, cells were overlaid with agarose, fixed with formalin, and stained with crystal violet. Plaque counts were used to calculate percentage neutralization and half-maximal inhibitory concentration (IC50) values. The experiments were carried out in duplicates.”
