The tumors were allowed to grow up to ~ 100–200 mm3, measured by calipers, after which mice received 0.5 mg/kg PSMA-1-Pc413 via tail-vein. To assess the in vivo transfection efficiency, 1 × 106 harvested B16F10 or B16F10-Luc-OVA cells were suspended in 100 μL PBS (pH 7.4) and injected subcutaneously into the left and right rear flanks of the female C57BL/6 mice (n = 1, 6−8 weeks old). R1-24 SNALP formulations with 50 μg of total pLuc and siLuc were intratumourally injected to B16F10 tumour-bearing mice and B16F10-Luc-OVA tumour-bearing mice on day 8 post-implantation, respectively. Each formulation was injected into two tumours in one mouse. One mouse was left untreated to serve as blank. D-luciferin potassium salt (Syd Labs Inc., Natick, MA) was dissolved in 1X PBS to a final concentration of 20 mg/ml. Mice were injected with D-firefly luciferin (150 mg/kg) subcutaneously and then sacrificed after 10 min, tumours were extracted and imaged to assess luminescence using an IVIS® Lumina III system (PerkinElmer, USA) at 6 h post-injection. Images were obtained using the bioluminescence mode with an auto exposure time, a binning factor of (HS)8, f number of 1.2, and a field of view of E−24 cm. Tumour images were analysed using Living Image v 4.7.3 software (PerkinElmer, USA), ROI was drawn around each tumour manually and the data is expressed as the total radiance per gram tumour for luciferase expression. The knockdown efficiency was calculated according to the following equation.
