大鼠抗小鼠PD-1单克隆抗体(克隆号RMP1-14,大鼠IgG2a kappa)可与由小鼠pdcd1基因编码的小鼠PD-1蛋白(CD279或程序性死亡-1)发生反应,该蛋白是免疫球蛋白超家族CD28家族的成员。PD-1有两个配体,即PD-L1和PD-L2,两者均属于B7家族。研究表明,在黑色素瘤小鼠模型中,通过抗小鼠PD-1抗体和抗小鼠PD-L1抗体进行治疗可以阻断PD-L1蛋白与其受体PD-1蛋白之间的相互作用,从而使肿瘤生长得到暂时性抑制。RMP1-14抗体能够阻断小鼠PD-L1蛋白和小鼠PD-L2蛋白与小鼠PD-1蛋白的结合。
我们的重组RMP1-14抗体包含大鼠抗小鼠PD-1抗体(杂交瘤克隆名称或编号:RMP1-14)的部分(可变区)或完整的氨基酸序列。这种鼠源化抗小鼠PD-1重组抗体不仅具有小鼠IgG抗体的恒定区,还具有鼠源化的可变区。
