该抗体结合仅存在于成熟T细胞和胸腺髓质细胞表面的T细胞受体-CD3(TCR-CD3)复合物中的CD3 epsilon(CD3ε)亚基。在体外实验中,T细胞、SP34-2与单核细胞之间的相互作用可诱导T细胞活化(有丝分裂)。另一方面,该抗体能阻断抗原受体与CD3之间的独特关联,并同时抑制细胞毒性T细胞的生成与功能。
首次给予该抗体后,T细胞会在数分钟至数小时内从血液循环中消失。在抗体治疗期间,表达常规表面分子组合(CD2、CD4和CD8)的T细胞会重新出现在外周血循环中,但这些细胞缺失CD3。停用抗体48小时后,包括CD3在内的完整表面分子组合会再次出现在所有T细胞表面。
通过内吞作用选择性移除CD3被认为是该抗体的主要作用机制。抗原受体与CD3的协同调制(共下调)解释了该抗体在体内的免疫阻断作用。如果CD3靶点分子未发生有效调制,该抗体在临床上将无法发挥疗效。
